What is papain?
Papain powder immature fruit contains papain, papa chymotrypsin A, papa chymotrypsin B, papaya peptidase B and other proteolytic enzymes. And the primary structure of four cysteine proteases is known to have a high degree of homology. Among them, papain is a thiol protease, which can hydrolyze the carboxyl ends of arginine and lysine in proteins and peptides, and can preferentially hydrolyze those peptides with two carboxyl groups or aromatic L-amino acids at the N-terminus of the peptide bond key.
Papain is a proteolytic enzyme with a molecular weight of 23406, consisting of a single peptide chain and containing 212 amino acid residues. At least three amino acid residues exist in the active center of the enzyme. They are Cys25, His159, and Asp158. When Cys25 is oxidized by an oxidant or bound to metal ions, the activity of the enzyme is inhibited, and the reducing agent cysteine (or Sulfite) or EDTA can restore enzyme activity. The other six cysteine residues form three pairs of disulfide bonds, none of which are in the active site.
